I travel into London on a regular basis, checking bus routes and train stations.
Piccadilly Circus is a nightmare for anyone who cannot climb stairs.
The closer stations in London are many of the main line stations such as Charing Cross, City ThamesLink, Liverpool street

Try the buses, you will be amazed. The only warning is that TFL often close bus stops for their own reasons. They are a bit rubbish at this.
I use the TFL site a lot to map the bus routes. The 24 and 29 both visit Trafalgar square and Leicester square along their routes.
If you fancy a bite to eat (some food), try the Crypt at St Martins in the Fields. The Crypt is an underground restaurant / cafe serving meals, soup, etc. to the weary traveller at a fair price. It is also very accessible, and has toilets.
OR get a cab, or an Uber.

Venue Access Information - copied from their official site on 1st December 2021:

There are three spaces for wheelchair users per performance, all located in the Royal Circle. Street level access is via Sherwood Street. There is one wheelchair space in Row A, and two in Box C (with companion seats), all located on the left hand side of the auditorium, Width of the entrance into Row A is 67.5cm, and the wheelchair space in Row A is 190cm wide and 95cm deep.
We can also accommodate transfer seating in Row A, seat 28 in the Royal Circle. Customers who wish to transfer should mention this at the time of booking?. If you need help transferring, we ask that you bring a companion. Please come to the Box Office on arrival and you will be shown the level access route to these seats. The auditorium opens 30 minutes before the performance.
Aisle seats and Row P in the Stalls have the most legroom. The Stalls bar is 22 steps down from the Foyer, the royal Circle Bar is 26 steps up from the Foyer, and it is 70 steps up to the Grand Circle Bar. ?Staircases are well lit and have handrails.

Accessibility :
The Piccadilly Theatre auditorium is equipped with a Sennheiser MobileConnect WiFi sound amplification system. MobileConnect uses WiFi to deliver superior, quality audio to a smart device such as an iPod or iPhone, either through headphones or via a necklace for hearing aid users. Devices can be borrowed on the day, or alternatively, patrons can download the Sennheiser MobileConnect app and use their own device.

Induction Loop Necklace :
Suitable for persons wearing a hearing aid, the induction loop necklace is worn around the neck. Whilst wearing the necklace, switch your hearing aid to the ‘T’ setting and the sound is amplified.

Headset :
Sound is amplified through headphones. Suitable for persons without a hearing aid. Guide, hearing and other working dogs are welcome in all parts of the theatre.
There is an accessible toilet next to Box C. Ladies and Gents toilets for Stalls seats are down 8 steps from the entrance to the auditorium. In the royal Circle, the Ladies toilets are up 8 steps from the bar, and the Gents toilets are down 15 steps from the bar level and then up a further 7. It is 70 steps up to the Grand Circle Bar, with the Ladies toilets on the way up to the bar, and the Gents toilet at the top.
